I used a Canon 1000d, with the Sigma 105mm macro, 65mm extension tubes, Raynox DCR-250 and Tumax combi flash (handheld, left hand brace technique)_____________________________________

I plan to buy Raynox DCR-250, but still dont know..
Is it good without 65mm extension tube and flash or? Can I use it without macro lens?

Yes of course you can use it with a normal lens 
you can also use it without a flash but I think a tripod is a duty
but without extensiontubes you canΒ΄t achieve this big magnification
