There is many articles about focus stacking technique, for example:

In my case, shooting that serie was pretty simple: i locked camera focus (using manual focusing mode, selected value in the middle of range and won't change it during shooting), move my macro setup, looking at camera screen, until i see frontal anthenna of the fly head in focus. Then i shoot 4 identical shots (for averaging them later), slightly move my macro setup forward, by fraction of millimeter, and locked focus moved forward. Then i shoot next 4 shots serie, move camera forward again, and so on, until i see that focus moved behing head of the fly. Then i upload all shots in computer, do RAW conversion of all of them with same parameters and averaged groups of 4 shots to make 16 images with different focus and lower noise after averaging. I load these 16 shots in Photoshop CS4 as layers, selected all of them and align (edit - auto align layers). Then i saved copy of aligned sources as PSD file and make focus stack (edit - auto blend layers, mode "stack images"). Result looks nice, but contains a lot of artifacts (blurred areas with bad focus). Then i load previously saved aligned sources on top of focus stacked layer and correct all these artifacts manually (by drawing masks on source layers). This was most time consuming part of whole processing. After artifact fixing, i do standard postprocessing (sharpening, noise removing, color correction, cropping).

Thanks! Yes, focus stack + averaging (16 groups of shots with different focus, each group contains 4 identical shots for averaging), 64 total.
