I try to use as little layers as possible, just the more of them the more confusing it can get (at least for me.) Most of the time I end up using four to five layers.
I start out with just a sketchy layer and if i really like it I will outline it on a new Pen layer (I use SAI )
And for the time, I guess it matters what im drawing really, Like Luigi here, it only took me about an hour I suppose.
My coloring style might look confusing, but it really isn't, its just a lot of scribbles of colors haha.
