Rainbow Dash growing a horn: I dunno, perhaps they'll all become alicorns 10 seasons from now, in which case it would work!
Pinkie Pie growing antlers: Perhaps an antler HAT, just for the Hearth's Warming episode? Or some kind of temporary antler-spell?
The Rarity and Applejack ones: Those could perhaps work in an episode where somebody's magic spells are going horribly wrong? (if the effect is temporary)
Fluttershy growing fangs: Yup, that's adorable, and what do you know, they actually found an excuse that made it work.
Spike getting spine-spines: Perhaps as Spike matures, they could show his growth by making his spines more jagged, which could be interpreted as spines on top of spines, therefore making this one work?

Don't you mean there is only one major change? Because these are obviously not spoilers. Twilight Sparkle becoming an alicorn is the only major change. The Mane Five and Spike's "changes" are false predictions. The Fluttershy fangs one was just a coincidence.
