That's not pipe smoke, it's actually ectoplasm. Here's a little more on his bio [link]
Essentially, he possesses his host by having them imbibe his floating ectoplasm through an object that was personal to him when he was alive (his pipe). Thereby granting him access to use their body.

So, the Inspectre is kind of like Hourman in that he takes pills to power him?

nope.
His alterego (the gentleman with the shotglass) is a separate personality. Inspectre is a ghost who posesses a human and their body takes on ghostly abilities.
