They are basically saying "We will never let you get a cutie mark" Because In the game they say "the curse had befallen on her this very night, she had the mark, she had to go!" so, what they mean about 'curse' and 'mark' is Getting a cutie mark; and "she" is Ruby the ghost you meet later, the pony you have been following the whole time. She's a ghost because...guess what they did to her.
